In photoshop CS5,
Draw shape with Rectangle tool ,
In layers panel , you will see two thumbnails shown...
double click on left thumbnails and pick white color ..
Then right click on this and choose blending options...
In Color Overlay tab .. Blend mode - Normal and color choose as you like ..
then You get transparent shape ..
